چکیده

The influence of pyridoxal-5'-phosphate on the activities of alanine aminotransferase and aspartate aminotransferase from commercial test sera was investigated. Enhancement of the activity of alanine aminotransferase by the coenzyme is about 20% when 0.1 mmol/1 of pyridoxal-S'-phosphate is added to the reaction medium. This concentration seems sufficient for full saturation of the apoenzyme. For aspartate aminotransferase, the enhancement of the enzymatic activity is much higher than for alanine aminotransferase and 0.1 mmol/1 of coenzyme is not sufficient for full saturation in this case.

Effects of temperature on measurement of aspartate aminotransferase and alanine aminotransferase in commercial control sera.

The catalytic activity concentrations of aspartate aminotransferase (EC 2.6.1.1) and alanine aminotransferase (EC 2.6.1.2) in several commercial control sera were determined at three different temperatures. Although these sera were of various biological origins, the temperature-conversion factors calculated are nearly identical to those for native human sera. The proportional increase of the ac...

Plasma pyridoxal 5'-phosphate concentrations in relation to apo-aminotransferase levels in normal, uraemic, and post-myocardial infarct sera.

The concentrations of pyridoxal 5'-phosphate, and the holoenzyme activities and apoenzyme contents of alanine aminotransferase and aspartate aminotransferase in plasma were determined simultaneously in healthy individuals, patients with renal insufficiency with and without chronic haemodialysis and in patients with acute myocardial infarction. Plasma pyridoxal 5'-phosphate is significantly dimi...

Two dry-reagent systems evaluated for determinations of enzyme activities.

We evaluated the Kodak Ektachem DT60/DTSC and the Boehringer Mannheim Reflotron for measuring activity concentrations of six enzymes. The Ektachem CVs for low concentrations of aspartate aminotransferase and alanine aminotransferase were high. As compared with the Ektachem and a routine "wet-chemistry" system, values for aspartate aminotransferase and alanine aminotransferase were lower as meas...

Clinical implications of differences between two recommended procedures for determination of aspartate aminotransferase.

We compared two officially recommended methods for determination of aspartate aminotransferase (EC 2.6.1.1): that of the International Federation of Clinical Chemistry (IFCC) and that of the Deutsche Gesellschaft für Klinische Chemie (DGKC). We used automated enzyme analyzers, initiating the reactions with 2-oxoglutarate. Normal values, 10-30 U/L (IFCC) and 7-18 U/L (DGKC), were apparently inse...

Decreased aminotransferase activity of serum and various tissues in the rat after cefazolin treatment.

Treatment of rats with cefazolin in vivo significantly suppressed activity of alanine and aspartate aminotransferases in serum and in the liver, brain, kidney, and heart. Simultaneous administration of pyridoxal further reduced enzyme activity except in the liver, where there was no change. Pyridoxal 5'-phosphate partly reversed the decreased enzyme activity in the serum, liver, and kidney, but...
